The ARE-Nrf2 Luciferase KeratinoSens™ Test Method is a sophisticated in vitro assay designed to evaluate the potential of various substances to activate the antioxidant response element (ARE) pathway, which is closely linked to the Nrf2 signaling pathway. This method is particularly relevant in the field of toxicology and skin sensitization testing, as it provides a reliable means of assessing the ability of chemicals to induce the expression of genes associated with the body's defense mechanisms against oxidative stress and inflammation.

In this assay, human keratinocyte cell lines are genetically modified to express a luciferase reporter gene under the control of the ARE. When a test compound is introduced, if it activates the Nrf2 pathway, Nrf2 translocates to the nucleus and binds to the ARE, leading to the transcription of the luciferase gene. The resulting luciferase enzyme can then be quantified through luminescence measurement, providing a clear and quantifiable indication of the compound's ability to activate the Nrf2 pathway.
